چکیده
Fig 6. Effect of Zn -enrichment on claudin-1 displacement rate. (A) NP-40-soluble and (B) NP-40insoluble fractions. Gray: calcium-free medium, black: Zn-enriched medium, white: Zn-enriched medium treated with STb for 6, 12 and 24 h. Lower panel: Immunoblot showing claudin-1 and GAPDH used to evaluate their relative amounts. NP-40 cell extracted proteins were separated on a 12% acrylamide SDSPAGE and immunoblotted with anti-claudin-1 and anti-GAPDH antibodies. The calcium-free medium was Zn -enriched (1.8 mM). There was no significant difference in claudin-1 dislogment rate under Znenriched condition compared to calcium-free medium. After 24 h, claudin-1 dislogment was observed as seen before in calcium-free medium (n = 3) (p<0.001). CLDN-1: claudin-1, GAPDH: Glyceraldehyde 3-phosphate dehydrogenase. Letters on top of the bars when different indicates a statistical difference between the treatments.

Escherichia coli STb Enterotoxin Dislodges Claudin-1 from Epithelial Tight Junctions
Enterotoxigenic Escherichia coli produce various heat-labile and heat-stable enterotoxins. STb is a low molecular weight heat-resistant toxin responsible for diarrhea in farm animals, mainly young pigs. A previous study demonstrated that cells having internalized STb toxin induce epithelial barrier dysfunction through changes in tight junction (TJ) proteins. These modifications contribute proba...
متن کاملEscherichia coli heat-stable toxin b impairs intestinal epithelial barrier function by altering tight junction proteins.
Escherichia coli heat-stable toxin b (STb) causes diarrhea in animals. STb binds to sulfatide, its receptor, and is then internalized. In the cytoplasm, through a cascade of events, STb triggers the opening of ion channels, allowing ion secretion and water loss and leading to diarrhea. Tight junctions (TJs) are well known for controlling paracellular traffic of ions and water by forming a physi...
متن کاملOuter Membrane Vesicles and Soluble Factors Released by Probiotic Escherichia coli Nissle 1917 and Commensal ECOR63 Enhance Barrier Function by Regulating Expression of Tight Junction Proteins in Intestinal Epithelial Cells
The gastrointestinal epithelial layer forms a physical and biochemical barrier that maintains the segregation between host and intestinal microbiota. The integrity of this barrier is critical in maintaining homeostasis in the body and its dysfunction is linked to a variety of illnesses, especially inflammatory bowel disease. Gut microbes, and particularly probiotic bacteria, modulate the barrie...
متن کاملLactobacillus rhamnosus strain GG prevents enterohemorrhagic Escherichia coli O157:H7-induced changes in epithelial barrier function.
Enterohemorrhagic Escherichia coli (EHEC) O157:H7 intimately attaches to intestinal epithelial monolayers and produces attaching and effacing (A/E) lesions. In addition, EHEC infection causes disruptions of intercellular tight junctions, leading to clinical sequelae that include acute diarrhea, hemorrhagic colitis, and the hemolytic-uremic syndrome. Current therapy remains supportive since anti...
متن کاملLipopolysaccharide Disrupts the Milk-Blood Barrier by Modulating Claudins in Mammary Alveolar Tight Junctions
Mastitis, inflammation of the mammary gland, is the most costly common disease in the dairy industry, and is caused by mammary pathogenic bacteria, including Escherichia coli. The bacteria invade the mammary alveolar lumen and disrupt the blood-milk barrier. In normal mammary gland, alveolar epithelial tight junctions (TJs) contribute the blood-milk barrier of alveolar epithelium by blocking th...
